We want to determine the aggregation landscape of TDP-43 in order to understand the contribution of different regions to aggregation.

At the same time, we want to understand the role of post-translational modifications (PTM)s of TDP-43 in the aggregation process because this seems to be a crucial and yet little-explored aspect of TDP-43 aggregation, and their effects could be dominant on the system.

The post holder will take advantage of our previous experience with ataxin-3 and other proteins to introduce ubiquitination, phosphorylation and glycation PTMs as have been observed in FTD/ALS patient tissues and will thus produce differently modified samples and characterize their RNA binding properties and the kinetics of aggregation with advanced biophysical methods including: circular dichroism, NMR, fluorescence, atomic force microscopy, electron microscopy and nanoIR.

Experience in biophysical techniques using advanced microscopy, NMR, biochemistry, and molecular biology approaches is required.

The successful applicants will join established research groups headed by Professor Annalisa Pastore.

This post will be offered on a fixed-term contract for 3 years.

This is a full-time post - 100% full time equivalent
