The project will evaluate the efficiency and cost-effectiveness of mutant nucleoside diphosphate kinase (mNDPK) systems towards the conversion of nucleoside diphosphates (NDPs) to triphosphates (NTPs). The candidate will be required to 1. Prepare mNDPK enzymes from a range of organisms (e.g. thermophiles); 2. Determine the breadth of scope of NDP substrates and the upper limits of mNDPK systems in terms of substrate concentrations and conversion rates; 3. Determine the limits of mNDPK systems in terms of longevity and thermal stability; 4 Explore immobilisation strategies, the performance of the enzyme system under continuous flow conditions, and the use of mNDPK systems in tandem with other key nucleotide-processing enzymes such as myokinase, which could facilitate AMP to ATP regeneration.
We are seeking an experienced Postdoctoral Researcher with expertise in the preparation, isolation and functional analysis of proteins. The primary technical responsibilities include undertaking protein production, protein activity assays, mostly by HPLC and substrate synthesis. In addition, translation of the technologies into a continuous flow chemistry must be undertaken. The post holder will analyse and interpret data, prepare written and oral reports on progress. They will also supervise PhD and MChem students in the laboratory and oversee routine maintenance on local instrumentation and liaise with equipment service professionals.
