RAF kinases play a major role in cancer, with BRAF being the most frequently mutated kinase in human cancers. For instance, 50% of malignant melanomas, 30-50% of thyroid and 15% of colorectal cancers are driven by mutated BRAF. Potent RAF inhibitors are in the clinic, but most patients become resistant while some tumour types with BRAF mutations respond poorly to start with. Interestingly, both acquired and intrinsic modes of RAF inhibitor resistance seem to be caused by network adaptations that allow the cancer cell to bypass the drug blockade. Using proteomic screens we have identified pathways that may mediate such bypasses and new pathways that may contribute to the oncogenic functions of RAF kinases. This project is already well progressed, and the postholder will focus on carrying out experiments that are needed to finish the project and prepare a publication. For this the postholder will work as part of an interdisciplinary team in Systems Biology Ireland.

Systems Biology Ireland (SBI, www.ucd.ie/sbi) has been consistently ranked amongst the top 5 systems biology research centres in the world by international review panels in 2014, 2016, and 2018. SBI has successfully developed a leading research programme focusing on the design of new diagnostic and therapeutic approaches to cancer based on a systems level, mechanistic understanding of cellular signal transduction networks. To accomplish these goals, SBI uses mathematical and computational modelling approaches in combination with cutting edge experimental technologies in genomics, transcriptomics, proteomics, advanced microscopy and flow cytometry as well as cell biology and molecular biology methods. The purpose-built SBI facility sits in the space between the UCD Health Sciences Centre and the UCD Conway Institute providing easy access to modern technology platforms, such as proteomics. SBI currently houses a multidisciplinary team of some 60 researchers including computational scientists and modellers, biologists, biochemists and clinician-scientists.
