Baxter Laboratory is seeking a Life Science Technician II (LST II).  This position will support various projects using CODEX technology in the laboratory of Dr. Garry Nolan.  Typical experiments will involve tissue sectioning, staining and running CODEX multidimensional staining protocol, antibody conjugation, operating robotic setups, mouse handling and dissection.

The LST II will perform immunofluorescent analysis of tissue sections and cell spreads, adjust the conditions for optimal performance of antibodies in tissue sections.

  • Will be involved with collecting, storing, and cutting tissues from HuBMAP and CRUK Consortia projects. This includes normal and cancerous tissues from the small intestine, colon, and esophagus. They will use the CODEX technology present in the Nolan lab to assist in the analysis of the tissue samples from the projects. CODEX imaging are specialized protocols for imaging, including use of a microfluidics robot. The LST II should be able to pay attention to great detail since there are multiple barcodes, many steps in the CODEX imaging pipeline, and many different unique biological reagents (antibodies/barcodes). This also requires lots of organization of reagents. The LST II will perform multiplexed CODEX imaging on such specimens and aid in data analysis.
  • Typical experiments will involve tissue sectioning, staining and running CODEX multidimensional staining protocol, antibody conjugation, operating robotic setups, mouse handling and dissection.
  • This position requires the ability to troubleshoot experimental assays in conjunction with other Nolan lab members.
  • The LST II will maintain and manage CODEX stocks/reagents (preparation of buffers, blocking reagents, inventory of oligo and antibodies, activating oligonucleotides, microscope components)
  • Conduct antibody-Oligo conjugations (using established protocols), testing antibody clones and oligo combinations to complete staining panels.
  • Troubleshoot antibody staining on various tissue types and clones to confirm positive and negative results.
  • Perform typical microscopy techniques such as H&E and fluorescent microscopy
  • Perform molecular biology protocols to verify antibody conjugations (PCR, Bradford Stains, SDS-page gels)
  • Perform multiplexed CODEX imaging using Keyence Microscope and Akoya Robot- become familiar with CODEX uploader software.
  • Aid in data analysis processing using established software, ImageJ, and some basic python code.
  • A standard laboratory notebook will be used to document daily activity.  Detailed protocols will be kept and updated as needed on Google Drive and other internal servers.

DESIRED QUALIFICATIONS:

College coursework in Biology, Biochemistry, Bioengineering, Biophysics, or in a related discipline.

Extensive prior experience with single-cell mass cytometry, fluorescence-based flow cytometry, characterization of cell lines. Excellent laboratory management skills and knowledge of the full range of genomic, proteomic, biochemical and immunohistochemical laboratory techniques.

Familiarity with and adherence to laboratory safety best-practices and Stanford University Health and Safety policies and regulations.

Working closely on a team as a member and as a leader on specific research projects is essential, sharing data, training colleagues and trainees in new techniques and protocols, documenting research protocols and results and recording and analyzing data is key.

The candidate will also assist with preparation of data, figures and reporting for manuscripts, progress reports and grant submissions: Proficiency in English and strong writing skills are desired.

EDUCATION & EXPERIENCE (REQUIRED):

Two years of laboratory experience or two years of college coursework in the scientific discipline a combination of equivalent education and relevant work experience.

KNOWLEDGE, SKILLS AND ABILITIES (REQUIRED):

Work at the Life Science Technician II level requires experience and ability beyond that required for the Life Science Technician I, including increased technical skills and problem-solving abilities

PHYSICAL REQUIREMENTS*:

  • Frequently stand, walk, twist, bend, stoop, squat, grasp lightly, use fine manipulation, grasp forcefully, perform desk-based computer tasks, use telephone, write by hand.
  • Ability to stand and move on hard surfaces for up to eight hours.
  • Occasionally lift and handle materials up to 10 pounds
  • Rarely carry/pull and/or push up to 20 pounds
  • Must have correctible vision to perform duties of the job.
  • Position may require repetitive motion.

WORKING CONDITIONS:

Work occurs in a research laboratory environment. Depending on research, may work in the vicinity of animals, chemicals, gasses, or hazardous materials. Expected to follow all safety requirements and procedures including wearing appropriate Personal Protective Equipment (PPE) if required.
